Intended use
This immunoassay kit allows for the in vitro quantitative determination of bovine Heat Shock Protein 90, HSP-90 concentrations in cell culture supernates, serum, plasma and other biological fluids.
 
Introduction
The 90 kDa heat shock protein, Hsp90, is one of the most abundant proteins in eukaryotic cells, comprising 1% of cellular proteins under non-stress conditions. Its contribution to various cellular processes including signal transduction, protein folding, protein degradation and morphological evolution has been extensively studied. The full functional activity of Hsp90 is gained in concert with other co-chaperones, playing an important role in the folding of newly synthesised proteins and stabilisation and refolding of denatured proteins after stress. Apart from its co-chaperones, Hsp90 binds to an array of client proteins, where the co-chaperone requirement varies and depends on the actual client.
 
Test principle
The microtiter plate provided in this kit has been pre-coated with an antibody specific to HSP-90. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for HSP-90 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain HSP-90, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of HSP-90 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
