Hi all:

I use Trizol to extract RNA from yeast. But it seems that I can not get any RNA but a lot of DNA
contamination.

Hope someone could help me.Wait for your answer.

Have a nice day!

I know of two reasons why this may happen with Trizol extraction. The first way to get DNA contamination is to homogenize the sample in too small a reagent volume- try adding more trizol to subsquent samples. Remember the sample volume should not exceed 10% of the volume of Trizol used for homogenization. The second is if the samples used for extraction contained organic solvents (ethanol, DMSO), strong buffers, or an alkaline solution. If this is the case you will need to 'clean up' your samples before proceeding with extraction. The protocol varies depending on what solvent is present in your samples.
