dear collegues,
i have a problem in ligation of 113bp into the second vector . after i cut from TA cloning with spe1 and BamH1 and purify from gel i got very small No. of ng usually self ligated plasmid or no colonies at all. the plasmid size around 4kb., i tried many molar ratio even 1 to 10 ( insert) but usually fail. if any one have advice please till me. thanks.
