    finding start and stop codon in an insert

    Hello molecular biology experts,

    This might be a stupid question to ask but, i don't know the answer for it and this is my first molecular biology experiment.

    Here is the story: Few days ago i got the bluescript vector with insert in it and i cultured the bacteria and prepared the plasmid and sequenced it. Then, i compared the sequence to bluescript vector first to find out where the junction is using vector NTI software. Later i compared the insert sequence to the already published sequence (down loaded from NCBI-Gene), for this i have used mRNA for the insert, is it right to use the mRNA for this purpose??

    Now my task is to find out the start and stop codon in my insert and to make virtual clone in an expression vector using Vector- NTI software. How do i find these codons?

    I hope you guys understand my explanation.

    Is there any forum or links or any books where i can get step by step explanation about how to make expression vector?

    I greatly appreciate your answers
