    Bacteria transformation

    Hellow all,
    A weird thing happened to me: I tranformed xl-1Blue bacteria with plasmid after being ligated by electrophoration. The bacteria was then seeded on LB agar plates with ampicilin, and the plate was incubated at 37 degree over night. Fortunately, I got about 8 colnes. However, when I picked thesee clones into LB medium with ampicillin, I didnt get any growth after incubation at 37 degree over night. It happened to me twice with the same ligation, but in other ligations and transformation performed in parallel everything was OK (meaning the colnes were grown in LB medium + Amp after being picked from the plate)

    Does somebody know what could be the problem? how I can overcome this problem
