hi,
i've been trying to point-mutate my 1.8 kb gene in 2.6 kb pUC19 using stratagene quick change kit but its simply not working even for the control, once i got colonies but no mutation , next time no colonies even in the control, switched to new kit, didn't work either, even in transformation of XL1 blue with pUC18, very few colonies, far less than expected according to the protocol.further i do see bands of amplified DNA on the gel, what could be wrong and where? any help will be greatly appreciated. thanks nbj
