    quick change site directed mutagenesis

    Hi,
    I am trying to change one nt in my protein already in a vector using the stratagene method. I have followed the stratagene method for my PCR, but have been unable to get product. I have tried increasing the template concentration and changing the extention time with no luck. My primers haven't been purified or phosphorylated, but the stratagene manual says this is not necessary for this method. I assume my primers are able to bind template as they have been sucessfully used in other PCR reactions. Any suggestions?
