    problems isolating BAC

    Dear all,

I have problems in isolating high amounts of bacterial artificial chromosomes (BAC`s). We just moved to a newe building leaving our ultra centrifuge behind since it was old, broken and not prepared anymore. Now I have to find a good protocol for doing miniprep using microfuges. The yield is ofcourse much lower but what can i do to increase this yield? What are the critical steps i have to take? And is it possible to seperate BAC`s from genomic E. coli DNA since it is moth circular DNA?
Thanks a lot for any suggestions.
    Nac
