Hi,
I was doing SSCP at 260 V, 12 Celsius degree overnight, and I do not have any problems. Now, I am trying to set up a protocol that needs to be run at 340V, the gel condition, and the temperature of the running are the same than my previous SSCP protocol. The first time that I run the gel, it came up ok, but after that I am having problem to get a good one. When I left the lab in the evening, the gel looks that it is going OK, but when I come back in the morning, the gel looks like has stretched up, and you can see part of the gel appear from the upper edge of the glass. After staining, the samples instead of being straight, are forming a curve shape making impossible of course to read the gel. What the problem could be? Any suggestion? Thanks,
