    Choice of restriction sites in MCS

    Hi

    Is it alright to use restriction sites that are next to each other in MCS for cloning? I am trying to subclone into an expression vector and I don't have much choice. The distance between the two restriction sites is only 1 nucleotide(huh!). Has anyone tried before?
