    cDNA-AFLP Fragment Reduction

    Dear All,

    My cDNA-AFLP fragment profiles only produce a low number of fragments (1 or 2 compared the the expected >20). This problem has only occured recently. Testing this proceudre on lambda DNA I have been able to determine that the banding pattern after the preamplification (no selection in the primers) has changed, which is what is causing the reduction in fragments numbers (I think?). I have tried all the standard things like making up fresh solutions but with no luck.

    Any suggestions would be appreciated.
