    I am trying to transfect a GFP-tagged protein into 293 cells. The first time I tried, I got very good transfection(>70%), on subsequent equations I got very variable transfection efficiencies - more often than not the transfection failed (<5%).

    Has anybody had a similar problem?

    We have tried with Qiagen's Effectene, superFect and Polyfect cell transfection reagent and have to say I am less than impressed. In parallel I also tested the much cheaper Gencarrier1 transfection reagent.

    Verdict: I just came back from the microscope and ~18 hours post-transfection (3T3 and 293 cells) I see tons of green cells with gencarrier1 and very few with superfect/polyfect. I’m often stick to brand name reagent. However, it @#$%& well better work the first time and the other times. I already have the system going and I’m not about to spend my time to troubleshoot an inconsistent transfection.
