    luciferase assay in HCV

    Hi,

    I'm currently working with the HCV replicon, trying a luciferase assay. It works pretty well, but when I compare my counts to the litterature, they're between 1-2 logs lower. Does anyone have a method for getting higher counts? Generally I work in 96W, though I did try 12W and then trypsinizing and lysing, but the results from that were so-so. On a similar note, has anyone heard of opaque welled plates in a format larger than 96W (ie 12, 24...) that could be used in a luminecence counter?
