HI, I want to know if it`s possible to add a sequence (of 30 nucleotides synthetised artificially) to an insert that I already had ligated in its vector. I have to subclone now into another vector ( pcDNA 3.1/Myc-HIs(+)plus) with the new sequence ligated to the insert. If someone has done it I would want to know if I can ligate directly or if I have to do something else.
