    effect of RNA on the process of southern blotting

    In the step of DNA preparation for southern blot analysis, is it necessary to treat DNA with RNase A before cut with restriction enzyme.

    I don't think it's necessary. mRNA are spliced products and will be degraded by RNase present in southern conditions. mRNA will not be cut by restriction enzymes even considering you are doing southern blot on a single exon gene. It shouldn't affect your southern result.
