    Digested DNA purified from agarose gel

    Hi, I am trying to purify digested genomic DNA (only select size of 19-22kb) from agarose gel, but I lost alot during the purification process. I am using QIAEX II Gel Extraction Kit. The purified DNA concentration is very low although I tried to decrease the final elution volume. So, is it any method to increase its concentration? I need the concentration up to around 500ng-1ug. Please help me if you guys know the way. Thanks.

    Ideally you would expect an recovery yield of around 75% for your large fragments. Did you use the fresh electrophoresis buffer? It would be worth to try using fresh buffers with correct pH. The major factor affects the beads binding is the pH. Good luck.
