    problem with DNA digestion

    I have some problem with the DNA digestion. I cut the DNA with HindIII restriction enzyme according to the reaction: 15 ug DNA, 100u enzyme, 1x buffer incubate at 37C for 16 hours. the result show the uncomplete cutting. Can anyone suggest me

    My suggestions:

    1. double check the DNA concentration. If it is genomic DNA it should not too viscous to do the digestion.

    2. double check the buffer compatibility. Use the digestion buffer chart and buffer from the same vendor unless you are sure the buffer compositions from different vendors are identical.
