    lambda HindIII marker

    why is it that the 4.4 kb band in the lambda Hind III marker is feinter than the bands below it? I heard that it has to do with the lambda genome/DNA itself but i can't seem to find any information about it. please help

    If the lamda DNA digestion is complete and is separated by 1-2% agarose gel it will show only eight bands. You can try to heat marker at 65 C for few minutes to disrrupt possible annealing between 4kb and 23kb bands which runs as extra 27kb band. If it is the case, it will decrease your intensity of your 4kb band.
