    SuperCos propagation

    Hello,

    Has anyone tried to propagate SuperCosI, the Cosmid from Stratagene (7.9kb)? I have tried electroporation into DH10B cells, but they don't grow very well - no colonies after O/N incubation, but then after I left the plate out on the bench O/N four colonies grow (pUC control had 4.9x10 9 CFU/ug). I tried to streak them onto new plates, but again I didn't get anything after O/N incubation at 37C.?
