dephosphorylation chew up

Hello,
I am having a slight problem constructing a cosmid library. Whenever I try to do a dephosphorylation there seems to be hardly any DNA left at the end. I used 0.1U CIAP/20pmol DNA. Therefore I run out of vector pretty fast, too. Now I was wondering whether there are any tricks not to loose all you DNA or whether I can grow some more of my 7.9kb cosmid by transforming it into the XL1-Blue cells or do I have to do an electroporation because it is too big? Any suggestions are very welcome. Thanks.

I searched the troubleshooting database using keywords dephosphorylation or CIP and it returned 41 hits. Hope it is helpful to you. Click for the result -> [biowww.net]
