alternative splicing

I got BAC clone for my interesting gene and I want to isolate entire promoter and gene to generate transgenic mice. However, I don't want to express the gene.
The reason why I want to put that gene along with promoter is that it helps the promoter efficienty. I am going to put GFP after promoter. If I make out of frame of the gene by adding one base pair of nucleotide right after GFP, is it possible that I can block the downstream of genes? Since there is two exons and one intron and the protein is a precursor which can be two other proteins by alternative splicing, I wonder making out of frame is good enough to block any translation of the genes. Need comments from experts. Thanks,
