Hello.
I'm trying to make different fragments of an interested gene region and clone them.
I researched the traditional exonucleaseIII procedure, but the gene is really short (~70 nt). Is there a different procedure available?

Just to synthesize the sequences (30-70 nt) and clone into your vector. No need for ExoIII digestion which is very hard to control enzyme conc. and timing with such short fragment.
