I am trying to do BrdU. I seeded the endothelial cells on 6 well plate. I got positive cells, but strangely, I usually found the positive cells in the peripheral area of the well but not in the centre. I don't understand why. This phenomenon affects the counting of positive cells. Can you tell me why? I followed the sigma protocol and shake all the reagents intermittently.
