Estradiol ELISA

WE have developed our own antibodies for estradiol using rabbits injected with Estradiol-CMO-BSA purchased from Steraloids. The test after three months revealed a good checkerboard response and the rabbits were bled accordingly. Since starting up the ELISA for the systen we have encountred problems in getting any response from the Standards. The checker board work well and gives positive results at various Antibody/ tracer dilutions. but just will not produce a standard curve with estradiol standards. WE have tried buying commercial tracer and we get the same results. Has anyone any ideas whre our problem lies before we go out and purchase the commercial estradiol antibody.?

Sounds like the problem is your standards. I think you may have to purchase them commercially.
