    Biotinylated Antibodies used in ELISA

    Hi all-
    I am working on developing a sandwich ELISA for the detection of Protein G in samples. I am using a commercially bought capture antibody that I biotinylate for use as the detection antibody. I just recieved a new maufacturer's lot of antibody and when I ran a standard curve to compare the old and new lots of detection (biotinylated) antibodies, the absorbance values of the new lot were much higher (about 10X) than the old. All of the conditions in the assays were identical except the detection antibodies. The antibodies are stored at 4C in PBS. Does anyone know if biotinylated antibodies lose potency or reactivity over time?
    Any suggestions are much appriciated.

    yes, the antibodies will lose potency overtime. it would be best to freeze them in useable aliqouts for long-term storage.
