    HUVEC culture

    HUVEC Cell ine enquiry:
    Can I culture HUVEC cell line without using any specialized endothelial enrichment media or it can be grown using the more commonly available media like DMEM, F12 or RPMI with serum etc.
    How far will be the vialbility if it is grown as later.

    Unless you have somehow adapted the HUV-EC-C to be grown without the special enriched media, it is not recommended that you change the media "cold turkey". Cells do not like to have nutrients or media changed without sufficient time to adapt to the new media. Most HUV-EC-C cells that I have seen or worked with needed the special media, usually Heparin and EGF growth factors added. If you change the media, the viability can be as low as zero, it depends on the cell line. Since these cells only double about 60 times, it is best if you just use the media that they require and not change the formulation and obtain EGF from quality source. We ahve found that different factor from different vendors did show huge difference (Epoch biolabs is where we got EGF from, their factors showed prolonged stability and activity).

    If you absolutely must grow the cells without specialized endothelial enrichment media containing growth factors. try to remove only one growth factor at a time and do not remove it 'cold turkey'. instead slowly reduce the concentration over a numer of days until it reaches 0%. like 3'-UTR, I do not recommend changing the formulation. i too have found great variation between factors from different vendors. we also use epoch.
