    help:how to choose a best RNAi vector

    hello,everyone.
    i have a problem, would you help me ? if i want to build up a RNAi vector system to transfect some stem cells,such as embryonic mesenchymal cell ,how to choose a best vector? Thank you very much,and look forward to your reply.

    There are some commercialy available siRNA vector systems you can chose from. Or you may want to construct your own shRNA vector. There is no such best vector since the silencing efficiency all depends on the specific shRNA you designed, the promoter (H1 or U6 etc) and transfection efficiency of your cell lines. The more efficient way of siRNA delivery is through virus vector such as lentivirus and adenovirus. Both are better choices for primary cells.
