    IPTG induction

    Could any body tell me why IPTG induction should be done at 20 C. Does it make any diiference if the cells were induced at higher tempreture

    Our lab constantly test IPTG induction under different temperatures because there is an optimum temperature for max expression for each protein. We've tested a range from 4oC to 37oC.

    I have cloned the gene of 1.2 Kb into pET vector. when inducing with IPTG or lactose i was not getting any expression.
    i tried lower temperatures,
    i tried diffrent IPTG and LActose concentrations.

    same gene was expressing under pBAD promoter?
    can u suggest me what to do to get expression under T7 promoter?
