    Protein aggregation

    Hi all,

I have a problem with this protein aggregation. I am just wondering whether anybody trouble shooted this during their work.

I m doing ammonuim sulphate precipitation, my protein tend to precipitate at 50% saturation, I dialyze it overnight with atleast two lt of buffer. After dialysis protein tends to aggregate and forms a kind of precipitate can any one tell me how to resolve this issue?

I have been asked to show the various stages of precipitation on SDS-PAGE, I have been using western to check the same. But I dont see much difference in the electrophoresis, does any one tell me do we really get to have big difference in precipitation, as per my knowledge goes, I cant really differentiate as other proteins will also get precipitated at the same saturation point. Any other suggestions?

    Thanks
    Geetha
