    PAGE on hydrophobic proteins

    Hi,

I am trying to run SDS PAGE on some artificial proteins synthesized in the lab. These proteins are very hydrophobic, so when I load them in the SDS gels, all I see is dark band in the wells after staining. I also performed western (these contain 10X His Tag) and see the same...for some reason these proteins are not flowing through the gels and are precipitating in the wells. I dialyze these proteins to remove Urea and freeze dry, the protein becomes completely insoluble so I am not able to characterize them either with some other techniques.

Is there a way that I can characterize these proteins or to see them on gels? Your help will really be appreciated.

    Thanks,
    Manoj

You can try tricine gel system (H. Schagger and G. von Jagow). It gives good resolution for hydrophobic proteins.
