    refolding problems, please help

    I am currently trying to refold a purified (somewhat) cysteine protease via dialyis. Dialyzing 6M urea from solution. 0.5M NaCl and 50mM monobasic sodium phosphate. pI of fusion protein is about 7.4, pH of buffer is 6.5. Once I get down to about 0.5M urea, when I go lower it crashes out of solution. Every time. i have tried going slower once I get to 0.5M (0.5 to 0.25 to 0M) and it still crashes. Does anyone have any suggestions?? I could use all the help I can get thanks
    I do have the luck of being able to re-solubilize the protein in 4M urea (dialysis) once it crashes.
