    Membrane insertion

    I am currently working on CLIC1, a chloride ion channel that exists in both soluble and integral membrane forms. My project focuses on the mechanisms and conditions that drive the soluble - membrane bound switch of CLIC1. Currently we are using isothermal titration calorimetry (ITC) to determine the binding thermodynamics of CLIC1 to liposomes (asolectin and sigma liposome formulation L4031: POPC, PGDO, cholesterol) and anionic detergent micelles (N-lauroylsarcosine). However, the ITC results can not distinguish between CLIC1 unfolding/rearrangement, binding and inserting into these artificial membranes. Hence, I need to create a system that will show CLIC1 membrane insertion and possibly identify its transmembrane domain (TMD). Could you please give me some ideas and references that can be used to demonstrate CLIC1 insertion and possibly identification of its TMD? Thanks for your time.
