    How much cells needed in the in vitro assay?

    Sorry to bother everyone, I have a question about in vitro ubiquitination assays. I guess my interested protein is an ubiquitin E3 ligase. so I want to use the protein IP from the transfected cells to be subjected to in vitro ubiquitination assay. I want use the IP protein instead of recombinant protein because the former protein matain the structural formation in the cells so the result is more reliable.But the problem is I don't how much cells I need for the assays?

    Would you please give me any clue? Thanks!!
