How to use MG132?

Hi, everyone:
In my experiment, I need to use MG132( a kind of proteasome inhibitor). I read some papers which all just provid the concerntration used on the cells, such as 10uM,but I still don't know the total amount used on the cells. For example, I treat the cells in the 10cm dish, how much I should use? Does anyone can help me? In addition, whether I should treat MG132 in SF medium or complete medium?? what the maximum hours I can use it on the cells?? Thanks!
