Cytochrome P450 quantification

i try to measure the amount of CYP P450 that i co-express with reductase in E coli using differential spectral method with CO (Omura and Sato, 1964). But i failed to get any positive reading. Anyone gives me some suggestions, I would highly appreciated. Thanks a lot!
