    protein extraction methadology

    Respected All,

    I am going to extract from plant material as well as seeds. I am using HEPES, DTT, Nacl and NaEDTA in extraction buffer in addition with aprotinin, chymostatin and leupeptin as protease inhibitor. I want to extract all protein from plant parts. I have two options:-
    1........ I use low amount of extraction buffer and then after centrifugation directly use it for SDS electrophoresis.
    2........ I use three to four time of extraction buffer and then precipitate protein by acetone and after that the pellet be used for SDS electrophoresis.
    which methodology will be suitable. The basic aim of my work to study total protein present in plant parts.
    Your kind suggestions/ guidance will be helpfull to me for batter work.
