    Scavenger Receptor western blot

    Hi!,
    We are performing the western blot of the Scavenger Receptor BI (SRBI) on hamster liver. We bougth an antibody against the carboxy terminus of the receptor, and we have found that the picture we obtained has a dark band near 150 kDa (just like in previous pictures of LDL receptor we obtained), and a less dark one near 75 KDa. The SRBI is supposed to appear at 82 kDa. Could anyone help us explaining what had happened?.

    Most probably your receprtor is post-translationaly modified by either proteolysis which may explain the 75 kDa band, and then froms dimers which may explain the 150 kDa band. If this is the case you need to find out the ratio between the monomer and dimer and this could tell more about the dynamics in relevance to the function of this protein at the stress you are studying However, contacting the antibody supplier with this data will help make the picture clearer.
