i am using bio-rad sequential extraction kit for extracting proteins from Moraxella catarrhalis for 2D SDS PAGE. It has 3 reagents for extraction of proteins depending on its solubility. I did get proetin spots in the first two gels run by first and second extraction reagents in the kit! Unfortunately, i did not get any spots for the third one!! Can u suggest of something !!

Strong horizontal stripes across the gel is almost reported as impurities in agarose overlay or equilibration solution. What other possible cause could be showing this symptom

It seems that you need to desalt your protein sample using desalting spin columns befor your run your sample on 2D gele electrophoresis. Check www.piercenet.com for 2d procedures as well.
