Dear sir,
I planned to work on comet assay, i dont find proper migration in slide. To 1/3rd of the slide i dipped hot normal melting agarose (1%) and dired(base slide) next day i loaded lymphocyte and Low melting agarose(5%)-2nd layer and finally 5% low melting agarose 80 micro litre addded placed in ice for 5 min.
Doubts:
1. How to place the slide in electrophoresis chamber
that is get near to anode or cathode???
2. We have to set the current both volts and mA or
either any one of it??????
3. How long i should boil the agarose both Normal
and low melting agarose. just allow them to dissolve or to the boiling condition???
