Hi i a, scraping J77A.1 cell line, when check the vabilty with trypan blue dilution 1:1 and 1:10 i am getting more (more then 50%) dead cells in 1:1 dilution when compared with 1:10 dilution,
i am following this protocol,
cell suspension (20 microliter) and 20 microliter of trypan blue loaded on heamocytometer,
1:10 dilution, 10 microliter cell suspension+90 microliter of trypan blue i didnt find a single dead cell, i am noting getting which method is correct, please suggest me,
