By Maria on 01-Mar-2002 I am performing a quantitation of the mRNA levels of several stress genes in a group of yeast strains, with RT and subsequent real time PCR. As standard samples (to construct the standard plot) I use always the same cDNA with serial dilutions. The problem: the efficiency of the PCR reaction and, hence, the slope of the standard curve varies from one experiment to another. So, I cannot compare the levels obtained in different experiments. If someone have ideas to solve this trouble, I would be most grateful. Thank you very much in advance. Rating: Good Reply
