Isotope-Coded Affinity Tag (ICAT) Analysis Introduction

An approach to high-throughput quantitative proteome analysis that comprises the following three components: (1) selective labeling of protein mixtures using isotope-coded affinity tag (ICATTM) reagents, (2) multidimensional chromatographic purification and separation of the peptide mixtures using a combination of analytical-scale strong cation exchange chromatography, avidin affinity chromatography, and microcapillary reverse-phase liquid chromatography, and (3) relative quantification and identification of the separated peptides by mass spectrometry.
