I am trying to establish stable mammalian cell lines
expressing a transfected gene by G418 selection in CHO
cells.
I treat the transfected cells with G418 @ 400ug/ml (I
assume this is this enough ?) but ... ...

I am trying to establish a stable mammalian cell line (hek293 cells) transfected with pcDNA3 vector, by using Geneticin as selective agent at 500 microgram/mL for a month or so, then at 200microgram/mL for maintaining. Cells have been growing well until recently they suddenly detached extensively after i splitted them just as usual.
That is really annoying. Could anybody tell me what is going wrong with my cells?
