This is a brief overview of how a Southern blot (more formally called an DNA blot) is performed and what type of data you can obtain form one. It includes a nice figure which shows how well the DNA digestion should be.

we are performing southern hybridization using genomic DNA of transgenic banana plants and gus probe. After restriction analysis we get very good smear but we only gets high mol. signals after hybridization. Signals are corresponding to about 16kb. please give some suggestion

I am not getting success in southern blotting as well as in hybridization too. my probe size is 136 bp. Is this size probe creates a problem in detecting the signa? if some one will help me with their better suggestion, i will greatful to him
