Why use PEG for transformation and single stranded DNA as a carrier Rating: n/a

Polyethylene glycol(PEG) have two effects on Ligation reaction for further transformation.
1.Increase the rate of ligation of blunt-ended DNA, allow ligations react at low concentration of enzyme and DNA. Ligation by PEG is highly depend on magnesium ions concentrations.
2.Alter the distribution of ligation products and suppress intramolecular ligation, keep all of DNA products are linear multimers.
