The double-immunofluorescence or triple-immunofluorescence technique uses two or three primary antibodies raised from hosts of different species. For example mouse-anti-actin and guinea pig-anti-integrin. Primary antibodies are mixed at pre-determined concentration before incubation with sections. It is pivatol to use highly specific secondary antibody in double-immunofluorescence staining to avoid cross-reaction and high background.
