By David Chang on 24-Aug-2006 Re:differences between traditional and multiplex PCR Multiplex PCR is to amplify several fragments at same reaction tube, up to eight primers can be used. It's good for saving time, and reagents. But The yield of each amplified product is reduced due to primer competettion. Before to set up multiplex PCR, to check fragments with each pair primers can be amplified efficiently in seperate reactions . Using Annealing Tm approximately primers. Increase primers concentration for ones reduced yield in multiplex reaction. And reduce amount the well-amplified primer pairs. Also increasing template amount if products poorly yield. generaly, in a multiplex reaction , a coulpe of primer pairs work poorly, you still can try touchdown PCR program.
