By Basha on 07-Dec-2007 i wanted to clone of 5.7 kb fragment in pGEMT easy vector.eventhough iam getting white colonies there was no insert only vector band was observed. my gene has NdeI at 5' end and hindIII at 3'end so i selected pET23a vector which has the same sites and ligated in 1:3 (vector:insert) but iam not getting waht may be the reason can u plz guide me to obtain clone of my gene of interest Rating: Very Good Reply
