Protein blocking solution for DNA hybridization
I routinely use the Boehringer Mannheim Blocking Reagent (sold as part of the
DIG system) as part of the prehybridisation/hybridisation mix when doing
32-P-labelled Northern and Southern blots, as this avoids the use of
(expensive, time consuming) denatured carrier DNAs such as herring sperm. The
UK catalogue doesn't say what this reagent is made of. Does anybody know? Are
there any alternative protein-based reagents (fat-free milk?) which can be used
as an alternative?

The blocking reagent consists of proteolytic fragments of casein in powder form ("The DIG system user's
guide for filter hybridization", Boehringer Mannheim). Casein is a phosphoprotein which occurs in milk
mainly in form of micelles.
