Content: Depending on the cell/tissue type of your smaples, you can chose from different protein lysis buffers. Commonly used protein lysis buffer includes NP-40, RIPA, Tris-triton etc. Lysis buffers differ in their ionic strength, detergent and its concentration.

RIPA (radioimmunoprecipitation assay buffer): more denaturing than NP-40 lysis buffer.

50mM Tris HCl, pH 8.0
150mM NaCl
1% NP-40
0.5% sodium deoxycholate
0.1% SDS

NP-40 (nonidet-P40) buffer:

50mM Tris HCl, pH 8.0
150mM NaCl
1% nonidet P-40

Commonly used protease inhibitors include:

- EDTA (chelated calcium and magnesium ions)
- Sodium orthovanadate (tyrosine protein phosphatases inhibitor)
- Sodium fluoride (serine/threonine protein phosphatases inhibitor)
- Aprotinin
- Leupeptin (lysosomal protease inhibitor)
- Pepstatin (aspartic proteases inhibitor)
- PMSF (protease inhibitor)

Some vendors also provide cocktails or tablets of protease inhibitors. (Sigma, Roche etc)
